Having a full-length cDNA transcript enables the investigation of a wide spectrum of gene functions through the application of key technologies.  It all starts with the full-length transcript.   On a larger scale, having all the gene transcripts in a biological pathway or gene family allows questions to be answered that could never before be addressed (Systems Biology).

Mammalian cell protein expression:  All OriGene clones are capable of driving the protein expression of each gene after transfection into mammalian cells.  Because all of the OriGene clones are driven by the same CMV-promoter, a uniform screening of groups of related genes is made possible.

 In vitro coupled transcription/translation:  The OriGene expression vector also supports coupled transcription-translation of the cDNA sequence using an appropriate cell-free system.  This approach may be used to generate recombinant proteins for testing activities in vitro and for target identifications.

siRNA/shRNA:  The preparation of dicer or RNaseIII products requires a full-length clone.  The isolation of naturally ocurring short hairpin (stem loop) structures also benefits from starting with full-length clones, especially those clones that contain untranslated flanking regions to the open reading frame.   It has been reported that some potent siRNA molecules against GFP are directed against these untranslated regions.

SNP detection:  Having a TrueClone transcript obtained directly from cDNA libraries removes the doubting question, "Is this nucleotide change a true polymorphism or was it artifactually created during PCR amplification?"  Once discovered, these SNPs can be evaluated and validated through patient studies.

Antibody production:  The OriGene clones can be engineered into tagged vectors to facilitate larger scale protein purification.  These proteins provide excellent antigens for antibody production and can be used in affinity purification. 

Hybridization probes:  There are multiple uses for nucleotide probes created from these plasmids including, Northern, southern, and chip hybridizations.

Access to splice variants:  During the quality control process, OriGene finds and collects alternatively spliced and other variant forms of each gene.  As our bioinformatics staff annotates these variants, each will be offered for sale and the variant "reports" will be available online for review prior to purchase.
