Background	CD8 is a 32kDa cell surface transmembrane glycoprotein expressed by most thymocytes, a subpopulation of mature peripheral blood T-cells and natural killer (NK) cells. CD8 is expressed on the cell surface as a heterodimer, comprising two distinct disulphide linked α and β polypeptide chains, or as a CD8α homodimer.   The CD8 αβ heterodimer is expressed on T-cell receptor (TCR) αβ+ cytotoxic T-lymphocytes (CTL’s), whereas CD8α homodimers are expressed on TCR γδ+ T-cells, natural killer cells and a subpopulation of TCR αβ+ T-cells. The CD8 molecule functions as a co-receptor (with the T-cell receptor) as part of the antigen-recognition complex on TCRαβ+, class I MHC-restricted T cells and plays an important role in T-cell signaling. The CD8 Ig-like domain binds to the alpha-3 domain of MHC-Class.
