Recommend Dilution | Sandwich ELISA: To detect Human sCD40 Ligand (using 100 μl/well antibody solution) a concentration of 0.5-2.0 μg/ml is required. In conjunction with Biotinylated Anti-Human sCD40-Ligand (Cat.-No AP01120BT-N or AP01120BT-S) as a detection antibody, it allows the detection of at least 0.2-0.4 ng/well of recombinant Human sCD40-Ligand. Western Blot: To detect Human sCD40-Ligand by Western blot analysis this antibody can be used at a concentration of 0.1-0.2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant Human sCD40-Ligand is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions. Immunohistochemistry on Paraffin sections: 2.5-5.0 µg/ml, Overnight at 4°C. This antibody stained sections of human cervical squamous cell carcinoma. An HRP-labeled polymer detection system was used with a DAB Chromogen. Heat induced antigen retrieval with a pH 6.0 Sodium citrate buffer is recommended. |

Background | CD40 ligand (CD40L) is a 33 kDa type II membrane glycoprotein expressed mainly on the cell surface of activated T lymphocytes, but also exists as a soluble form extracellularly. CD40L is the ligand for CD40, a member of the TNF superfamily, which is expressed on the cell surface of B cells, macrophages/monocytes, dendritic cells, vascular endothelial cells, and epithelial cells. CD40L plays an important role in B cell proliferation, antibody class switching, modulation of apoptosis in the germinal center through interaction with B cells expressing CD40, and activation of CD4+ T cells. Mutation within the CD40L gene is linked to hyper IgM syndrome, an X linked immunodeficiency disease that is characterized by elevated level of serum IgM and decreased level of other isotypes. |
