Recommend Dilution	Immunohistochemistry on Paraffin Sections: 2.0 µg/ml with an overnight incubation at 4°C. Staining of Formalin-Fixed, Paraffin Embedded Sections of Human breast invasive ductal carcinoma using an HRP labeled polymer detection system with a AEC Chromogen (Heat induced antigen retrieval with a pH 6.0 Sodium Citrate buffer is recommended). Neutralization: To yield one-half maximal inhibition (ND50) of the biological activity of Human RANTES (100 ng/ml), a concentration of 0.4-0.6 µg/ml of this antibody is required. Sandwich ELISA: To detect Human RANTES by Sandwich ELISA (using 100 µl/well antibody solution) a concentration of 0.5-2.0 µg/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotinylated Anti-Human RANTES (Cat.-No PP1064B1 or PP1064B2) as a detection antibody, allows the detection of at least 0.2-0.4 ng/well of recombinant Human RANTES. Western Blot: To detect Human RANTES use a concentration of 0.1-0.2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant Human RANTES is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.

Background	Rantes, also known as CCL5, is the protein classified as a chemotactic cytokine or chemokine. This protein is a chemotactic for blood monocytes, memory T helper cells, eosinophils and basophils, and plays an active role in recruiting leukocytes into inflammatory sites. Rantes also induces the proliferation and activation of certain natural-killer (NK) cells to form CHAK (CC-Chemokine-activated killer) cells. It is also an HIV-suppressive factor released from CD8+ T cells.
