Summary | Transcriptional activator that binds to the consensus sequence 5'-AACAAAG-3' in the promoter of target genes and plays an essential role in embryonic cardiovascular development and lymphangiogenesis (PubMed:7651823, PubMed:10742113, PubMed:12748961, PubMed:18931657, PubMed:19429912, PubMed:26939885). Activates transcription of PROX1 and other genes coding for lymphatic endothelial markers (PubMed:18931657, PubMed:26939885). Plays an essential role in triggering the differentiation of lymph vessels, but is not required for the maintenance of differentiated lymphatic endothelial cells (PubMed:18931657). Plays an important role in postnatal angiogenesis, where it is functionally redundant with SOX17 (PubMed:16895970). Interaction with MEF2C enhances transcriptional activation (PubMed:11554755). One of the four constructs at minimum are guaranteed to produce 70% or more gene expression knock-down provided a minimum transfection efficiency of 80% is achieved. Western Blot data is recommended over qPCR to evaluate the silencing effect of the shRNA constructs 72 hrs post transfection. To properly assess knockdown, the gene expression level from the included scramble control vector must be used in comparison with the target-specific shRNA transfected samples.
