Bioactivity | Specific: . One unit will oxidize 1.0 umol of UDP-glucose to UDP-glucuronic acid per minute at pH 8.0 at 25°C. Activity Assay 1. Prepare a 1470 ul assay buffer. The final concentrations are 93mM Tris-Hcl (pH 8.0), 0.2mM UDPG, and 1mM beta-NAD. 2. Add 20 ul of recombinant protein UGDH with 1 ug, 2 ug and 5 ug in assay buffer 3. Mix by inversion and load 200 ul of reaction mix in to a plate well. 4. Record the decrease in A340nm for 5 minutes at 25°C. |

Summary | The protein encoded by this gene converts UDP-glucose to UDP-glucuronate and thereby participates in the biosynthesis of glycosaminoglycans such as hyaluronan, chondroitin sulfate, and heparan sulfate. These glycosylated compounds are common components of the extracellular matrix and likely play roles in signal transduction, cell migration, and cancer growth and metastasis. The expression of this gene is up-regulated by transforming growth factor beta and down-regulated by hypoxia.
