Bioactivity | Specific: > 1.5 units/ml. One unit will convert 1.0 umole of 2 - phosphoglycerate to phospho(enol)pyruvate per minute at pH 7.5 at 25°C. Activity Assay: 1. Prepare 1,450 ul assay buffer. The final concentrations are 81 mM Triethanolamine, 1.9 mM 2 - phosphoglycerate, 0.12 m M beta - NADPH , 25 mM magnesium sulfate, 100 mM potassium chloride, 1.3 mM ADP, 4 unit pyruvate kinase , 6 unit L - lactic dehydrogenase. 2. Add 50 ul of recombinant beta - enolase protein in various concentrations ( 0.25 ug, 0.1 ug, 0.5 ug ) in assay buffer. 3. Mix by inversion and load 200 ul of reaction mix in to a plate well . 4. Record the decrease in A 340 nm for 5 minutes at 25°C. |

Summary | This gene encodes one of the three enolase isoenzymes found in mammals. This isoenzyme is found in skeletal muscle cells in the adult where it may play a role in muscle development and regeneration. A switch from alpha enolase to beta enolase occurs in muscle tissue during development in rodents. Mutations in this gene have be associated glycogen storage disease.
