Recommend Dilution	Neutralization To yield one-half maximal inhibition [ND50] of the biological activity of mRANTES (100.00 ng/ml), a concentration of 6.0-8.0 µg/ml of this antibody is required. ELISA Indirect: To detect mRANTES by indirect ELISA (using 100 µl/well antibody solution) a concentration of 0.5-2.0 µg/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with compatible secondary reagents, allows the detection of 0.2-0.4 ng/well of recombinant mRANTES. Sandwich: To detect mRANTES by Sandwich ELISA (using 100 µl/well antibody solution) a concentration of 0.5-2.0 µg/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotinylated Anti mRantes (PP1081B1 or PP1081B2) as a detection antibody, allows the detection of at least 0.2-0.4 ng/well of recombinant mRANTES. Western Blot To detect mRANTES by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant mRANTES is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.

Background	RANTES is a chemoattractant for blood monocytes, memory T helper cells and eosinophils. It causes the release of histamine from basophils and activates eosinophils. It binds to CCR1, CCR3, CCR4 and CCR5 and is one of the major HIV suppressive factors produced by CD8+ T cells. Recombinant RANTES protein induces a dose-dependent inhibition of different strains of HIV 1, HIV 2, and simian immunodeficiency virus (SIV). It is T cell and macrophage specific, induced by mitogens, and belongs to the intercrine beta (chemokine CC) family.
