Bioactivity | DNMT1 activity verified in a biochemical assay: DNMT1 (DNA (cytosine-5-)-methyltransferase 1) (TP326414) is a key methyltransferase that is responsible for maintaining methylation patterns established in development. DNMT1 preferentially methylates hemi-methylated CpG di-nucleotides and associates with DNA replication sites in S phase maintaining the methylation pattern in the newly synthesized strand. Varying concentrations of DNMT1 were added to a microplate containing a bound methyltransferase substrate. After incubation, the resulting methylated DNA residues were detected immunologically and a colorimetric signal was generated and measured. |

Summary | This gene encodes an enzyme that transfers methyl groups to cytosine nucleotides of genomic DNA. This protein is the major enzyme responsible for maintaining methylation patterns following DNA replication and shows a preference for hemi-methylated DNA. Methylation of DNA is an important component of mammalian epigenetic gene regulation. Aberrant methylation patterns are found in human tumors and associated with developmental abnormalities. Variation in this gene has been associated with cerebellar ataxia, deafness, and narcolepsy, and neuropathy, hereditary sensory, type IE. Alternative splicing results in multiple transcript variants.
